The invention provides an isolated inflammation-related gene, the protein molecule encoded by the gene, and the use of the gene in diagnosis and treatment of inflammatory conditions.
DNA sequences which are capable of converting L-malate into L-lactate are incorporated into suitable vectors and used to transform both prokaryotic and eukaryotic hosts. The DNA sequences and vectors are particularly useful in conferring the ability to perform malolactic fermentation on wine producing yeasts. Saccharomyces cerevisiae 2514-10C/ySWl and E. coli RRI/ySWl were deposited at the A.T.C.C. on Aug. 16, 1982, and given accession Ser. Nos. 20651 and 39176, respectively.
Hybrid DNA capable of effecting controlled amplification in a host cell of a heterologous gene that determines a specific function. The hybrid DNA has the following three different regions of DNA: (1) an “amplicon” that is inducible by an external stimulus; (2) the heterologous gene (or a site for its insertion); and (3) a selectable marker enabling isolation of the transformed host cells. In order to make a desired compound coded by the heterologous gene, a vehicle including the hybrid DNA is i…
A Salmonella gene, encoding a cytolysin, has been identified by screening for hemolysis on blood agar. The gene (slyA) is present in every strain of Salmonella examined in Shigella, and enteroinvasive Escherichia coli (EIEC) but not in other enterobacteriaceae. It is encoded near 28.5 minutes on the chromosome. A SlyA (salmolysin) has hemolytic and cytolytic activity and has a molecular weight predicted by the DNA sequence. LD.sub.50 and infection kinetics data in mice indicate that the toxin is…
A technique suitable for cloning a cDNA having a base sequence coding for the ACTH/LPH precursor is disclosed. The invention is exemplified by the cloning of a cDNA fragment comprising a base sequence coding for the endorphin region. The fragment, hereinafter termed the endorphin gene cDNA sequence, was obtained from cultured mouse pituitary tumor cells known to produce the ACTH/LPH precursor protein.
The present invention relates to somatic mutations in the Multiple Tumor Suppressor (MTS) gene in human cancers and their use in the diagnosis and prognosis of human cancer. The invention further relates to germ line mutations in the MTS gene and their use in the diagnosis of predisposition to melanoma, leukemia, astrocytoma, glioblastoma, lymphoma, glioma, Hodgkin’s lymphoma, CLL, and cancers of the pancreas, breast, thyroid, ovary, uterus, testis, kidney, stomach and rectum. The invention also…
Methods and compositions are provided for gene transfer to intact mammals with expression of the exogenous genetic material in the host. Mammalian host cells which are regenerative, normally highly proliferative or subject to induced proliferation, are transformed or modified in vitro with DNA capable of replication and expression in the host cell, wherein the DNA becomes incorporated into the cell. The modified cells are found to regenerate in the host with expression of the introduced DNA. Par…
This invention is a process to produce specific proteins coded for by eukaryotic (or prokaryotic) DNA in bacteria. The invention, which uses recombinant DNA techniques, produces proteins in their natural, functional state unencumbered by extraneous peptides.
Double stranded DNA (ds-DNA) can be prepared by preparing a hybrid DNA containing a single stranded portion and a double stranded portion and carrying out in vivo gap repair on the hybrid DNA. The hybrid DNA is prepared by synthesizing a single strand of DNA and introducing the single strand into double stranded DNA. This method of synthesis can be used to make synthetic genes, including synthetic DNA coding for the TAT protein of HIV-I, and incorporates useful restriction sites. Also included a…
A transacting DNA binding factor is disclosed. The ASF-1 protein factor specifically binds to the sequence motif TGACG found upstream of the promoter in many plant genes. Co-expression of this protein factor augments the level of expression of the up-regulated promoter containing the TGACG motif.
